RESUMO

Sawmill workers are exposed to wood dust (a well-known carcinogen), microorganisms, endotoxins, resin acids (diterpenes), and vapours containing terpenes, which may cause skin irritation, allergy, and respiratory symptoms including asthma. The health effects of most of these exposures are poorly understood as most studies measure only wood dust. The present study assessed these exposures in the Norwegian sawmill industry, which processes predominantly spruce and pine. Personal exposures of wood dust, resin acids, endotoxin, fungal spores and fragments, mono-, and sesquiterpenes were measured in 10 departments in 11 saw and planer mills. The geometric mean (GM) and geometric standard deviation (GSD) thoracic exposures were: 0.09 mg m-3 dust (GSD 2.6), 3.0 endotoxin units (EU) m-3 (GSD 4.9), 0.4 × 105 fungal spores m-3 (GSD 4.2), 2 × 105 fungal fragments m-3 (GSD 3.2), and 1560 ng m-3 of resin acids (GSD 5.5). The GM (GSD) inhalable exposures were: 0.72 mg m-3 dust (2.6), 17 EU m-3 (4.3), 0.4 × 105 fungal spores m-3 (3.8), and 7508 ng m-3 (4.4) of resin acids. The overall correlation between the thoracic and inhalable exposure was strong for resin acid (rp = 0.84), but moderate for all other components (rp = 0.34-0.64). The GM (GSD) exposure to monoterpenes and sesquiterpenes were 1105 µg m-3 (7.8) and 40 µg m-3 (3.9), respectively. Although mean exposures were relatively low, the variance was large, with exposures regularly exceeding the recommended occupational exposure limits. The exposures to spores and endotoxins were relatively high in the dry timber departments, but exposures to microbial components and mono-and sesquiterpenes were generally highest in areas where green (undried) timber was handled. Dust and resin acid exposure were highest in the dry areas of the sawmills. Low to moderate correlation between components (rp ranging from 0.02 to 0.65) suggests that investigations of exposure-response associations for these components (both individually and combined) are feasible in future epidemiological studies.

Reactive oxygen species, potentially formed through environmental exposures, can overwhelm an organism's antioxidant capabilities resulting in oxidative stress. Long-term oxidative stress is linked with chronic diseases. Pesticide exposures have been shown to cause oxidative stress in vivo. We utilized a longitudinal study of corn farmers and non-farming controls in Iowa to examine the impact of exposure to the widely used herbicides atrazine and 2,4-dichlorophenoxyacetic acid (2,4-D) on markers of oxidative stress. 225 urine samples were collected during five agricultural time periods (pre-planting, planting, growing, harvest, off-season) for 30 farmers who applied pesticides occupationally and 10 controls who did not; all were non-smoking men ages 40-60. Atrazine mercapturate (atrazine metabolite), 2,4-D, and oxidative stress markers (malondialdehyde [MDA], 8-hydroxy-2'-deoxyguanosine [8-OHdG], and 8-isoprostaglandin-F2α [8-isoPGF]) were measured in urine. We calculated ß estimates and 95% confidence intervals (95%CI) for each pesticide-oxidative stress marker combination using multivariate linear mixed-effect models for repeated measures. Farmers had higher urinary atrazine mercapturate and 2,4-D levels compared with controls. In regression models, after natural log transformation, 2,4-D was associated with elevated levels of 8-OHdG (ß = 0.066, 95%CI = 0.008-0.124) and 8-isoPGF (ß = 0.088, 95%CI = 0.004-0.172). 2,4-D may be associated with oxidative stress because of modest increases in 8-OHdG, a marker of oxidative DNA damage, and 8-isoPGF, a product of lipoprotein peroxidation, with recent 2,4-D exposure. OBJECTIVES: The aims were to study biomarkers of systemic inflammation, platelet/endothelial activation and thrombosis in tunnel construction workers (TCW). METHODS: Biomarkers and blood fatty acids were measured in blood of 90 TCW and 50 referents before (baseline) and towards the end (follow-up) of a 12 days work period. They had been absent from work for 9 days at baseline. Air samples were collected by personal sampling. RESULTS: Personal thoracic air samples showed geometric mean (GM) particulate matter and α-quartz concentrations of 604 and 74 µg/m3, respectively. The arithmetic mean (AM) concentration of elemental carbon was 51 µg/m3. The GM (and 95% confidence interval) concentration of the pro-inflammatory cytokine TNF-α decreased from 2.2 (2.0-2.4) at baseline to 2.0 pg/mL (1.8-2.2) (p = 0.02) at follow-up among the TCW. Also the platelet activation biomarkers P-selectin and CD40L decreased significantly [25.4 (24.1-26.6) to 24.4 (22.9-26.0)] ng/mL, p = 0.04 and 125 (114-137) to 105 (96-115) pg/mL, p < 0.001, respectively. ICAM-1 concentrations increased from 249 (238-260) to 254 (243-266) ng/mL (p = 0.02). No significant alterations were observed among the referents when assessed by paired sample t test. Unbeneficial alterations in blood fatty acid composition were observed between baseline and follow-up, mainly among referents. CONCLUSIONS: TCW had slightly reduced systemic inflammation and platelet activation although highly exposed to particulate matter, α-quarz and diesel exhaust, which might be due to increased physical activity during the exposure period. The slightly increased ICAM-1 may indicate monocyte recruitment to the lungs. The diet was substantially altered towards a less beneficial fatty acid profile.

BACKGROUND: Aluminum potroom exposure is associated with increased mortality of COPD but the association between potroom exposure and annual decline in lung function is unknown. We have measured lung volumes annually using spirometry from 1986 to 1996. The objective was to compare annual decline in forced expiratory volume in 1 s (dFEV1) and forced vital capacity (dFVC). METHODS: The number of aluminum potroom workers was 4,546 (81% males) and the number of workers in the reference group was 651 (76% males). The number of spirometries in the index group and the references were 24,060 and 2,243, respectively. RESULTS: After adjustment for confounders, the difference in dFEV1 and dFVC between the index and reference groups were 13.5 (P < 0.001) and -8.0 (P = 0.060) ml/year. CONCLUSION: Aluminum potroom operators have increased annual decline in FEV1 relative to a comparable group with non-exposure to potroom fumes and gases.

3-Hydroxy acids are constituents of the lipid A part of lipopolysaccharides and may potentially be used as chemical markers of endotoxin. While commercial enzymatic assays, such as the widely used Limulus amebocyte lysate (LAL) assay, commonly detect merely the water-soluble fraction of the bioactive endotoxin, the chemical approach aims to estimate the total amount of endotoxin present in a sample. Our objective was to develop a simple method for quantitative profiling of 3-hydroxy fatty acids in occupational and environmental samples based on detection with HPLC-MS/MS. We included eleven 3-hydroxy fatty acids (3-hydroxyoctanoic acid to 3-hydroxyoctadecanoic acid) in the HPLC-MS/MS based method, which involved base hydrolysis of filter samples using 1M sodium hydroxide and removal of the base as well as concentration of the fatty acids using solid-phase extraction on a functionalized polystyrene-divinylbenzene polymer. Recovery trials from spiked glass fiber filters, using threo-9,10-dihydroxyhexadecanoic acid as internal standard, gave an overall recovery of 54-86% for 3-hydroxy fatty acids of medium chain length (3-hydroxynonanoic to 3-hydroxypentadecanoic acid). 3-Hydroxyoctanoic acid and the longer chain fatty acids were more problematic yielding overall spike recoveries of 11-39%. While the 3-hydroxy fatty acid profile of pure lipopolysaccharides was dominated by 3-hydroxydecanoic, 3-hydroxydodecanoic and 3-hydroxytetradecanoic acid the aqueous phase from drilling mud contained in addition relatively high amounts of 3-hydroxyoctanoic and 3-hydroxynonanoic acid. Endotoxin activity as measured by the LAL assay was reasonably correlated (R(2)=0.54) to the sum of 3-hydroxydecanoic acid, 3-hydroxydodecanoic acid and 3-hydroxytetradecanoic acid in these samples.

BACKGROUND: The Norwegian aluminum industry developed and implemented a protocol for prospective monitoring of employees' exposure using personal samplers. We analyzed these data to develop prediction lines to construct a job exposure matrix (JEM) for the period 1986-1995. METHODS: The protocol for personal monitoring of exposure was implemented in all seven Norwegian aluminum plants in 1986 and continued until 1995. Personal samplers were used to collect total dust, fluorides, and total polycyclic aromatic hydrocarbons (PAH). In addition, exposure could be categorized according to process, i.e. prebake, Søderberg, and 'other'. We constructed four-dimensional JEMs characterized by: Plant, Job descriptor, Process, and Year. Totally 8074, 6734, and 3524 measurements were available for dust, fluorides, and PAH, respectively. The data were analyzed using linear mixed models with two-way interactions. The models were assessed using the Akaike criterion (AIC) and unadjusted R (2). The significance level was set to 10% (two-sided) for retaining variables in the model. RESULTS: In 1986, the geometric mean (95% confidence interval in parentheses) for total dust, total fluorides, and PAH were 3.18 (0.46-22.2) mg m(-3), 0.58 (0.085-4.00) mg m(-3), and 33.9 (2.3-504) µg m(-3), respectively. During 10 years of follow-up, the exposure to total dust, fluorides, and PAH decreased by 9.2, 11.7, and 14.9% per year, respectively. Each model encompassed from 49 to 72 significant components of the interaction terms. The interaction components were at least as important as the main effects, and 65 to 91% of the significant components of the interaction terms were time-dependent. CONCLUSION: Our prediction models indicated that exposures were highly time-dependent. We expect that the time-dependent changes in exposure are of major importance for longitudinal studies of health effects in the aluminum industry.

OBJECTIVES: Little is known about the working conditions and airway inflammation in hairdressers in Palestine. We aimed to investigate if hairdressers in Palestine have a higher level of airway inflammation as compared to a control group. We also assessed the hairdressers' physical working conditions and exposure to ammonia gases at the hair salons. Lastly, we investigated the association between ammonia levels and inflammation markers in the airways and the blood. METHODS: Our study participants were 33 non-smoking hairdressers (aged 19-50 years) and 35 non-smoking control subjects (aged 18-49 years). Both groups answered a questionnaire on respiratory symptoms, and performed lung function and exhaled nitric oxide (eNO) tests. Blood and sputum samples were collected from all participants and air concentration levels of ammonia were measured in 13 salons. RESULTS: Hairdressers had a higher level of sputum neutrophil count (absolute numbers/mg sputum (median (25th-75th centiles)) compared to controls, 376 (183-980) and 182 (96-358), respectively. Hairdressers also had significantly elevated eNO and blood C reactive protein (CRP) levels compared to the control subjects, controlled for age and body mass index. Exposure measurements showed that the hairdressers in salons with scarce ventilation were exposed to ammonia concentration, ranging from 3 to 61 mg/m(3). CONCLUSIONS: Compared to unexposed controls, the hairdressers had signs of neutrophilic airway inflammation, higher eNO levels and higher CRP. The hairdressers were exposed to high concentrations of ammonia from hairdressing chemicals and their working conditions were unsatisfactory.

PURPOSE: The aim was to study inflammatory biomarkers in tunnel construction workers (TCW). METHODS: Surfactant protein D (SP-D), Clara cell protein 16 (CC-16) and C-reactive protein (CRP) were studied in serum of 90 TCW and 50 referents before and at the end of an 11-day work period. Personal air sampling was carried out on the two consecutive days before follow-up. RESULTS: The TCW's geometric mean exposure to particulate matter and α-quartz were 604 and 74 µg/m(3), respectively. The arithmetic mean concentration of elemental carbon was 51 µg/m(3). The arithmetic mean concentration of SP-D was reduced by 7.6 µg/L in the TCWs and 0.6 µg/L in the referents (p = 0.04) at the end as compared to before the work period. Subjects who had ever been TCW had lower arithmetic mean CC-16 concentrations at baseline (5.4 µg/L) than subjects who had never worked as TCW (6.4 µg/L). Years worked as TCW was significantly associated with an annual mean decline of the CC-16 concentration of 0.04 µg/L. The concentrations of the biomarker of systemic inflammation, CRP, were not affected by exposure in the TCWs. Current smoking and body mass index have a large impact on the measured biomarker concentrations. CONCLUSIONS: The results suggest that former and current TCWs have lower serum CC-16 concentrations than referents, while the concentrations of SP-D decreased during exposure. The serum biomarker of systemic inflammation, CRP, was not altered during exposure. Current smoking and BMI were related to the concentrations of all measured biomarkers.

OBJECTIVES: Tunnel construction workers are exposed to particulate and gaseous air contaminants. Previous studies carried out in the 1990s showed that tunnel construction workers were at increased risk of both short-term and long-term lung function decline. Since then, efforts have been made to reduce exposure. The objective of the present study was to investigate if current exposure may still cause short-term lung function impairment. METHODS: Tunnel workers work 12 days consecutively, and then they are off for 9 days. Ninety tunnel workers and 51 referents were examined with spirometry and questionnaires before their work period started and again 11 days later. Personal exposure to particles and α-quartz in the thoracic aerosol subfraction, elemental carbon and organic carbon, oil mist, nitrogen dioxide and ammonia was assessed on two consecutive days between the two health examinations. RESULTS: The geometric means air concentrations for particulate matter in the thoracic mass aerosol subfraction, α-quartz, oil mist, organic carbon and elemental carbon for all workers were 561, 63, 210, 146 and 35 µg/m(3), respectively. After 11 days of work, the mean forced expiratory volume in 1 s (FEV1) in healthy participants had declined 73 mL (SD 173), p<0.001 in the tunnel workers, compared to 3 mL (SD 21), p=0.9 in the referents. Also, forced vital capacity (FVC) had declined significantly. Declines in FVC and FEV1 were significantly associated with exposure to organic carbon. CONCLUSIONS: In spite of reduced levels of exposure in modern tunnelling operations, a negative impact on lung function was still observed.

OBJECTIVES: Personal air measurements of aerosols and gases among tunnel construction workers were performed as part of a 11-day follow-up study on the relationship between exposure to aerosols and gases and cardiovascular and respiratory effects. METHODS: Ninety tunnel construction workers employed at 11 available construction sites participated in the exposure study. The workers were divided into seven job groups according to tasks performed. Exposure measurements were carried out on 2 consecutive working days prior to the day of health examination. Summary statistics were computed using maximum likelihood estimation (MLE), and the procedure NLMIXED and LIFEREG in SAS was used to perform MLE for repeated measures data subject to left censoring and for calculation of within- and between-worker variance components. RESULTS: The geometric mean (GM) air concentrations for the thoracic mass aerosol sub-fraction, α-quartz, oil mist, organic carbon (OC), and elemental carbon (EC) for all workers were 561, 63, 210, 146, and 35.2 µg m(-3), respectively. Statistical differences of air concentrations between job groups were observed for all contaminants, except for OC, EC, and ammonia (P > 0.05). The shaft drillers, injection workers, and shotcreting operators were exposed to the highest GM levels of thoracic dust (7061, 1087, and 865 µg m(-) (3), respectively). The shaft drillers and the support workers were exposed to the highest GM levels of α-quartz (GM = 844 and 118 µg m(-3), respectively). Overall, the exposure to nitrogen dioxide and ammonia was low (GM = 120 and 251 µg m(-) (3), respectively). CONCLUSIONS: Findings from this study show significant differences between job groups with shaft drilling as the highest exposed job to air concentrations for all measured contaminants. Technical interventions in this job should be implemented to reduce exposure levels. Overall, diesel exhaust air concentrations seem to be lower than previously assessed (as EC).

BACKGROUND: This study examines the possible effect of exposure to aerosols and gases on lung function in a fertilizer plant in Norway. METHODS: Dynamic lung volumes (FVC and FEV1) of 383 workers were measured in 2007 and 2010. During the follow-up period, most workers performed tasks with low exposure levels of acid aerosols and inorganic gases. The overall median inhalable and thoracic aerosol exposure levels were 1·1 and 0·21 mg/m(3), respectively. A questionnaire on respiratory symptoms was provided. RESULTS: During the follow-up period, there was an adjusted decrease of FEV1 of 18 ml/year (P<0·001). The respiratory symptoms score was low during follow-up. CONCLUSIONS: Work in this fertilizer industry may lead to an excessive lung function decline. We have, however, not been able to find any plausible exposure related explanation for the overall lung function decline.

INTRODUCTION: The aim of this study was to identify important determinants of dust exposure in the Norwegian silicon carbide (SiC) industry and to suggest possible control measures. METHODS: Exposure to total dust, respirable dust, quartz, cristobalite, SiC, and fiber was assessed in three Norwegian SiC plants together with information on potential determinants of exposure. Mixed-effect models were constructed with natural log-transformed exposure as the dependent variable. RESULTS: The exposure assessment resulted in about 700 measurements of each of the sampled agents. Geometric mean (GM) exposure for total dust, respirable dust, fibers, and SiC for all workers was 1.6mg m(-3) [geometric standard deviation (GSD) = 3.2], 0.30mg m(-3) (GSD = 2.5), 0.033 fibers cm(-3) (GSD = 5.2), and 0.069mg m(-3) (GSD = 3.1), respectively. Due to a large portion of quartz and cristobalite measurements below the limit of detection in the processing and maintenance departments (>58%), GM for all workers was not calculated. Work in the furnace department was associated with the highest exposure to fibers, quartz, and cristobalite, while work in the processing department was associated with the highest total dust, respirable dust, and SiC exposure. Job group was a strong determinant of exposure for all agents, explaining 43-82% of the between-worker variance. Determinants associated with increased exposure in the furnace department were location of the sorting area inside the furnace hall, cleaning tasks, building and filling furnaces, and manual sorting. Filling and changing pallet boxes were important tasks related to increased exposure to total dust, respirable dust, and SiC in the processing department. For maintenance workers, increased exposure to fibers was associated with maintenance work in the furnace department and increased exposure to SiC was related to maintenance work in the processing department. CONCLUSION: Job group was a strong determinant of exposure for all agents. Several tasks were associated with increased exposure, indicating possibilities for exposure control measures. Recommendations for exposure reduction based on this study are (i) to separate the sorting area from the furnace hall, (ii) minimize manual work on furnaces and in the sorting process, (iii) use remote controlled sanders/grinders with ventilated cabins, (iv) use closed systems for filling pallet boxes, and (v) improve cleaning procedures by using methods that minimize dust generation.

The aim of this study was to characterise personal exposures to dust, acid vapours, and gases among workers in a Norwegian nitrate fertiliser production plant, as part of an ongoing epidemiological study. In total, 178 inhalable and 179 thoracic aerosol mass fraction samples were collected from randomly chosen workers (N = 141) from three compound fertiliser departments (A, B and C), a calcium nitrate fertiliser production department, nitric acid- and ammonia-production departments, and a shipping department. The overall median inhalable and thoracic aerosol mass concentrations were generally low (1.1 mg m(-3) (min-max: <0.93-45) and 0.21 mg m(-3) (min-max: <0.085-11), respectively). Workers at the compound fertiliser departments B and C had significantly higher inhalable aerosol mass air concentrations compared to the other departments (p < 0.05), except for compound fertiliser department A; however, the difference between the compound fertiliser department C and calcium nitrate department was slightly above the significant level. Workers at the compound fertiliser department A had significantly higher thoracic aerosol mass air concentrations compared to the other departments (p < 0.05), except for compound fertiliser departments B and C. The results indicate that the extrathoracic aerosol fraction of the aerosol compared to the thoracic fraction dominated in most departments. Measurement of the main constituents Ca, K, Mg, and P in the water-soluble and water-insoluble aerosol mass fractions showed that the air concentrations of these elements were low. There is, however, a shift towards more water-soluble species as the production goes from raw material with phosphate rock towards the final product of fertilisers. Overall, the arithmetic mean of water-soluble Ca in the thoracic mass fraction was 51% (min-max: 1-100). A total of 169 personal samples were analysed for HNO(3) vapour and HF. The highest median concentration of HNO(3) (0.63 mg m(-3)) was in the compound fertiliser departments B, and all measurements but four of the HF concentrations were below the LOD of 190 µg m(-3). Exposures to NH(3), CO and NO(2) were measured using direct-reading electrochemical sensors and the time weighted overall averages were all below the LODs of the respective sensors, NH(3) 2 ppm; CO 2 ppm; and NO(2) 0.2 ppm, but some short-term peaks were detected. Even though our results indicate that the workers may experience peak exposure episodes when performing job tasks such as cleaning or maintenance work, the overall air concentrations are well below what is considered to cause known health risks.

The objective was to construct a retrospective job-exposure matrix (JEM) for the Norwegian silicon carbide industry. More than 3300 historical total dust measurements were available (1967-2005); however, there were few measurements of other agents. Total dust measurements were therefore used as the basis for the JEM, and a novel method was developed to estimate exposure to other agents. Multiple linear regression models were developed to describe historical exposure to total dust. Exposure estimates were extrapolated backward to periods without exposure data by adjustments for process and work-hour related changes. An exposure assessment study was performed where total dust was sampled in parallel with fibers or respirable dust. The respirable dust was analyzed for the content of quartz, cristobalite, and silicon carbide. Mixed-effect models were developed to estimate the exposure to these agents from total dust exposure, plant, and job group. Exposure to asbestos and polycyclic aromatic hydrocarbons was assigned qualitatively. Multiple linear regression models of total dust described historical exposure best in the furnace department (R(2) (adj) = 0.49-0.74). Models in the other departments explained less variance (R(2) (adj) = 0.12-0.32). Exposure determinants and total dust explained a substantial proportion of the between- (70-100%) and within-worker (8.0-54%) variance in the mixed-effect models. The relative bias between available historical measurements and the estimated exposure to dust components varied between -39% (fiber) and 40% (quartz). However, corrections were not considered necessary due to limitations in the historical data. The component-specific metrices were sufficiently different from each other (r(Pearson) < 0.7), with the exception of total and respirable dust (r(Pearson) = 0.84) and total dust and cristobalite (r(Pearson) = 0.72), and will enable component-specific epidemiologic analyses in the future. Improved and less correlated estimates of exposure levels for the different components in the dust were obtained with the updated exposure assessment. BACKGROUND: Previous epidemiologic findings suggest an association between exposure to trichloroethylene (TCE), a chlorinated solvent primarily used for vapor degreasing of metal parts, and non-Hodgkin lymphoma (NHL). OBJECTIVES: We investigated the association between occupational TCE exposure and NHL within a population-based case-control study using detailed exposure assessment methods. METHODS: Cases (n = 1,189; 76% participation rate) and controls (n = 982; 52% participation rate) provided information on their occupational histories and, for selected occupations, on possible workplace exposure to TCE using job-specific interview modules. An industrial hygienist assessed potential TCE exposure based on this information and a review of the TCE industrial hygiene literature. We computed odds ratios (ORs) and 95% confidence intervals (CIs) relating NHL and different metrics of estimated TCE exposure, categorized using tertiles among exposed controls, with unexposed subjects as the reference group. RESULTS: We observed associations with NHL for the highest tertiles of estimated average weekly exposure (23 exposed cases; OR = 2.5; 95% CI, 1.1­6.1) and cumulative exposure (24 exposed cases; OR = 2.3; 95% CI, 1.0-5.0) to TCE. Tests for trend with these metrics surpassed or approached statistical significance (p-value for trend = 0.02 and 0.08, respectively); however, we did not observe dose­response relationships across the exposure levels. Overall, neither duration nor intensity of exposure was associated with NHL, although we observed an association with the lowest tertile of exposure duration (OR = 2.1; 95% CI, 1.0-4.7). CONCLUSIONS: Our findings offer additional support for an association between high levels of exposure to TCE and increased risk of NHL. However, we cannot rule out the possibility of confounding from other chlorinated solvents used for vapor degreasing and note that our exposure assessment methods have not been validated.

Methods for measurements and the potential for occupational exposure to organophosphates (OPs) originating from turbine and hydraulic oils among flying personnel in the aviation industry are described. Different sampling methods were applied, including active within-day methods for OPs and VOCs, newly developed passive long-term sample methods (deposition of OPs to wipe surface areas and to activated charcoal cloths), and measurements of OPs in high-efficiency particulate air (HEPA) recirculation filters (n = 6). In total, 95 and 72 within-day OP and VOC samples, respectively, have been collected during 47 flights in six different models of turbine jet engine, propeller and helicopter aircrafts (n = 40). In general, the OP air levels from the within-day samples were low. The most relevant OP in this regard originating from turbine and engine oils, tricresyl phosphate (TCP), was detected in only 4% of the samples (min-max

The aim was to develop quantitative estimates of farmers' pesticide exposure to atrazine and to provide an overview of background levels of selected non-persistent pesticides among corn farmers in a longitudinal molecular epidemiologic study. The study population consisted of 30 Agricultural Health Study farmers from Iowa and 10 non-farming controls. Farmers completed daily and weekly diaries from March to November in 2002 and 2003 on pesticide use and other exposure determinants. Urine samples were collected at 10 time points relative to atrazine application and other farming activities. Pesticide exposure was assessed using urinary metabolites and diaries. The analytical limit of detection (LOD) ranged between 0.1 and 0.2 microg/l for all pesticide analytes except for isazaphos (1.5 microg/l) and diazinon (0.7 microg/l). This article describes a systematic review of the industrial hygiene literature for uses of trichloroethylene (TCE) in industry for the exposure assessment of two population-based case control studies of brain cancer in the United States. Papers and reports that address uses of and exposures to TCE were identified from MEDLINE, TOXLINE, NIOSHTIC, the NIOSH Health Hazard Evaluation database (keywords: chlorinated solvents and trichloroethylene), and in other reviews. This search was complemented by reviewing the reference lists from the identified literature. The collected information was systematized by the Standard Industrial Classification (SIC) system, and measurement data reported in the literature were summarized in a database. TCE use was extensive from the early 1920s through the 1970s mainly as a degreasing agent in metal-fabricating operations. After the 1970s it became less popular because of environmental concerns. TCE historically has had a multitude of uses in many other industries, e.g., dry cleaning, textile, electronics, leather, and rubber. Also, many products like adhesives, drugs, paints, inks, and various industrial products have contained TCE. It was banned as a food additive and in cosmetics in 1977. The arithmetic mean (AM) of the measurements across all industries and decades was 38.2 ppm. The highest personal and area air levels were reported in vapor degreasing (AM of 44.6 ppm). Most TCE measurements were performed in the 1950s, 1970s, and 1980s. The data described here could be used by exposure assessors as is to identify the presence and approximate levels of exposure. Using the same information as a basis should increase the reliability of the assessments, making it easier to compare both the exposure assessment methods and the epidemiologic results across different studies.

Elimination of interfering substances in urine by solid phase extraction (SPE) prior to analysis resulted in 10-fold improvement in the sensitivity of atrazine mercapturate (AM) enzyme-linked immunosorbent assay (ELISA) compared to previous reports. Of the two tested SPE systems, Oasis HLB and MCX, the mixed-mode MCX gave good recoveries (82%) of AM in spiked samples measured by ELISA, whereas the reverse-phase HLB phase was not compatible with the immunochemical method. At relatively high concentrations of urinary AM (>20 ng mL(-1)), sample dilution was effective enough for the elimination of interfering substances. The new liquid chromatography-mass spectrometry (LC-MS) method developed for AM utilizes online-SPE with Oasis HLB, column switching and a stable-isotope internal standard. The limit of quantification (0.05 ng mL(-1)) indicates improved sensitivity compared with most previously published LC-MS methods for AM. Validation of all three methods, LC-MS, ELISA+SPE and ELISA+dilution with spiked urine samples showed good correlation between the known and measured concentrations with R2 values of 0.996, 0.957 and 0.961, respectively. When a set (n=70 plus 12 blind duplicates) of urine samples from farmers exposed to atrazine was analyzed, there was a good agreement (R2=0.917) between the log normalized data obtained by ELISA+SPE and LC-MS. High correlation among the data obtained by the two tested methods and the LC-MS method by the Center of Disease Control and Prevention (CDC), together with low variability among the blind duplicates, suggests that both methods reported here would be suitable for the analysis of urinary AM as a biomarker for human exposure of atrazine.

In tunnel construction work, dust is generated from rock drilling, rock bolting, grinding, scaling, and transport operations. Other important dust-generating activities are blasting rock and spraying wet concrete on tunnel walls for strength and finishing work. The aim of this study was to identify determinants of dust exposure in tunnel construction work and to propose control measures. Personal exposures to total dust, respirable dust, and alpha-quartz were measured among 209 construction workers who were divided into 8 job groups performing similar tasks: drill and blast workers, shaft drilling workers, tunnel boring machine workers, shotcreting operators, support workers, concrete workers, outdoor concrete workers, and electricians. Information on determinants was obtained from interviewing the workers, observation by the industrial hygienist responsible for the sampling, and the job site superintendent. Multivariate regression models were used to identify determinants associated with the dust exposures within the job groups. The geometric mean exposure to total dust, respirable dust, and alpha-quartz for all tunnel workers was 3.5 mg/m(3) (GSD = 2.6), 1.2 mg/m(3) (GSD = 2.4), and 0.035 mg/m(3) (GSD = 5.0), respectively. A total of 15 percent of the total dust measurements, 5 percent of the respirable dust, and 21 percent of the alpha-quartz exceeded the Norwegian OELs of 10 mg/m(3), 5 mg/m(3), and 0.1 mg/m(3), respectively. Job groups with highest geometric mean total dust exposure were shotcreting operators (6.8 mg/m(3)), tunnel boring machine workers (6.2 mg/m(3)), and shaft drilling workers (6.1 mg/m(3)). The lowest exposed groups to total dust were outdoor concrete workers (1.0 mg/m(3)), electricians (1.4 mg/m(3)), and support workers (1.9 mg/m(3)). Important determinants of exposure were job group, job site, certain tasks (e.g., drilling and scaling), the presence of a cab, and breakthrough of the tunnel. The use of ventilated, closed cabs appeared to be the single most important control measure for lowering exposures.
